Derivados de Imidazotiadiazol

Antecedentes de la invenciïn La invenciïn tenïa por objeto hallar nuevos compuestos con valiosas propiedades, en particular aquellas que se pueden utilizar para la preparaciïn de medicamentos.

La presente invenciïn hace referencia a compuestos y al uso de compuestos en los que la inhibiciïn, la regulaciïn y/o la modulaciïn de la transducciïn de seïales por parte de las quinasas, en particular de los receptores de quinasas de TGF–beta, desempeïa un papel, ademïs de a composiciones farmacïuticas que comprenden estos compuestos, y al uso de los compuestos para el tratamiento de enfermedades inducidas por quinasas.

El factor de crecimiento transformante beta es el prototipo de la superfamilia TGF–beta, una familia de factores de crecimiento pleiotrïficos altamente conservados, que llevan a cabo importantes funciones tanto durante el desarrollo embrionario, como tambiïn en el organismo adulto. En los mamïferos se han identificado tres isoformas de TGF– beta (TGF–beta 1, 2 y 3) , siendo TGF–beta 1 la isoforma mïs frecuente (Kingsley (1994) Genes Dev 8:133–146) . TGF–beta 3 se expresa, por ejemplo, sïlo en cïlulas mesenquimales, mientras que el TGF–beta 1 se encuentra en cïlulas mesenquimales y epiteliales. El TGF–beta se sintetiza como una preproproteïna y se libera en una forma inactiva en la matriz extracelular (Der y nck (1985) Nature 316: 701-705; Bottinger (1996) PNAS 93: 5877-5882) . Ademïs de la prorregiïn separada, a la que tambiïn se denomina pïptido asociado a latencia (LAP, por sus siglas en inglïs) y que queda asociada con la regiïn madura, una de las 4 isoformas de las proteïnas de uniïn a TGF–beta latente (LTBP 1–4) puede tambiïn unirse a TGF–beta (Gentr y (1988) Mol Cell Biol 8: 4162–4168, Munger (1997) Kindey Int 51: 1376–1382) . La activaciïn del complejo inactivo que es necesaria para el desarrollo de la acciïn biolïgica del TGF–beta aïn no se ha aclarado completamente. Sin embargo, es sin duda alguna necesario un procesamiento proteolïtico, por ejemplo mediante plasmina, transglutaminasa plasmïtica o trombospondina (Munger (1997) Kindey Int 51: 1376–1382) . El ligando activado TGF–beta actïa de mediador en su acciïn biolïgica a travïs de tres receptores TGF–beta en la membrana, los receptores de tipo I y de tipo II de expresiïn ubicua y los receptores de tipo III betaglicano y endoglina, siendo el ïltimo expresado ïnicamente en cïlulas endoteliales (Gougos (1990) J Biol Chem 264: 8361–8364, Loeps–Casillas (1994) J Cell Biol 124:557–568) . Ambos receptores TGF–beta del tipo III carecen de un dominio quinasa intracelular que facilita la transmisiïn de seïales a la cïlula. Debido a que los receptores TGF–beta de tipo III se unen con gran afinidad a las tres isoformas TGF–beta, y tambiïn a que el receptor TGF–beta de tipo II posee una gran afinidad por los ligandos unidos al receptor de tipo III, se cree que la funciïn biolïgica consiste en la regulaciïn de la disponibilidad de los ligandos para los receptores TGF–beta de tipo I y de tipo II (Lastres (1996) J Cell Biol 133:1109–1121; Lopes–Casillas (1993) Cell 73: 1435– 1344) . Los receptores de tipo I y de tipo II estrechamente relacionados estructuralmente, poseen un dominio serina/treonina quinasa, que es responsable de la transmisiïn de seïales, en la regiïn citoplasmïtica. El receptor TGF–beta de tipo II se une al TGF–beta, tras lo cual el receptor TGF–beta de tipo I es reclutado para este complejo de transmisiïn de seïales. El dominio serina/treonina quinasa del receptor de tipo II es constitutivamente activo y puede fosforilar radicales de serilo en este complejo en el asï llamado dominio GS del receptor de tipo I. Esta fosforilaciïn activa la quinasa del receptor de tipo I, que ahora es por sï mismo capaz de fosforilar los mediadores de seïales intracelulares, las proteïnas SMAD, y de ese modo inicia la transmisiïn de seïales intracelulares (resumido en Der y nck (1997) Biochim Biophys Acta 1333: F105–F150) .

Las proteïnas de la familia SMAD sirven como sustratos para todas las quinasas receptoras de la familia de TGF– beta. Hasta ahora se han identificado 8 proteïnas SMAD, las cuales pueden ser divididas en 3 grupos: (1) las SMAD asociadas a receptores (R–SMAD) son sustratos directos de las quinasas receptoras de TGF–º (SMAD1, 2, 3, 5, 8) ;

(2) Las co–SMAD, que se asocian con los R–Smad durante la cascada de seïales (SMAD4) ; y (3) las SMAD inhibidoras (SMAD6, 7) , las cuales inhiben la actividad de las proteïnas SMAD mencionadas con anterioridad. De las diferentes R–SMAD, las SMAD2 y SMAD3 son los mediadores de seïales especïficos de TGF–beta. En la cascada de seïales TGF–beta, las SMAD2/SMAD3 son por tatno fosforiladas por el receptor TGF–beta del tipo I, lo que les permite asociarse con SMAD4. El complejo de SMAD2/SMAD3 y SMAD4 resultante se puede translocar ahora en el nïcleo celular, donde puede iniciar la transcripciïn de los genes regulados por TGF–beta directamente o a travïs de otras proteïnas (resumido en Itoh (2000) Eur J Biochem 267: 6954–6967; Shi (2003) Cell 113: 685–700) .

El espectro de las funciones de TGF–beta es muy amplio y depende del tipo celular y del estado de dife-renciaciïn (Roberts (1990) Handbook of Experimental Pharmacology: 419–472) . Las funciones celulares que se encuentran influenciadas por TGF–beta incluyen: apoptosis, proliferaciïn, diferenciaciïn, movilidad y adhesiïn celular. Por consiguiente, el TGF–beta desempeïa un papel importante en una variedad muy amplia de procesos biolïgicos. Durante el desarrollo embrionario, se expresa en sitios de morfogïnesis y en particular en ïreas con interacciïn epitelial–mesenquimal, en las que induce importantes procesos de diferenciaciïn (Pelton (1991) J Cell Biol 115:1091–1105) . El TGF–beta realiza tambiïn una funciïn clave tambiïn en la autorrenovaciïn y mantenimiento de un estado no diferenciado de cïlulas madre (Mishra (2005) Science 310: 68-71) . Ademïs, el TGF–beta tambiïn lleva a cabo importantes funciones en la regulaciïn del sistema inmune. En general ejerce una acciïn inmunosupresora,

ya que inhibe, entre otros, la proliferaciïn de linfocitos y restringe la actividad de macrïfagos tisulares. El TGF–beta permite de este modo que las reacciones inflamatorias disminuyan nuevamente y por tanto ayuda a evitar reacciones inmunolïgicas excesivas (Bogdan (1993) Ann NY Acad Sci 685: 713-739, resumido en Letterio (1998) Annu Rev Immunol 16: 137-161) . Otra funciïn del TGF-beta es la regulaciïn de la proliferaciïn celular. El TGF-beta inhibe el crecimiento de cïlulas de origen endotelial, epitelial y hematopoyïtico, pero promueve el crecimiento de cïlulas de origen mesenquimal (Tucker (1984) Science 226:705-707, Shipley (1986) Cancer Res 46:2068–2071, Shipley (1985) PNAS 82: 4147-4151) . Otra funciïn importante del TGF-beta es la regulaciïn de la adhesiïn celular y las interacciones cïlula-cïlula. El TGF-beta promueve la formaciïn de la matriz extracelular mediante la inducciïn de proteïnas de la matriz extracelular, tales como, por ejemplo, fibronectina y colïgeno. Ademïs, TGF-beta reduce la expresiïn de metaloproteasas degradantes de la matriz e inhibidores de las metaloproteasas (Roberts (1990) Ann NY Acad Sci 580: 225-232; Ignotz (1986) J Biol Chem 261: 4337-4345; Overall (1989) J Biol Chem 264: 1860-1869) ; Edwards (1987) EMBO J 6: 1899-1904) .
